15A and 50A circuit in 1 conduit.

Aside from de-rating, and wire fill, what other issues should I consider.

My application will start at a ?J? box, run #4 wires through flex aluminum with 50A protection feeding a sub-panel then running a 15A 12 gauge branch circuit back to the ?J? box through the same conduit.

I prefer to keep the branch circuits away from main feeders but I cant think of any code requiring the same.
 
No rule to prohibit branch and feeders in same raceway.
Flex Al- do you meet requirements for EGC in 250.118?
Remember you adjust ampacity for fill from the 90 C rating, 12 THHN is 30 amps, typically can't exceed 20 amps,but see 240.4(D) and (B)

The insulation systems of all conductors must be rated for the highest class of voltage represented within the conduit. But that is generally not an issue for most of what we do, since most of our conductors are rated for 600 volts, and most of our equipment is either 120 or 480 volts.
 
One other "issue" I would consider is that once you've reached the #4 AWG sized conductor you'll need to follow the 6X and 8X rules for box sizing.
