With this exception about the entries or exits coming into the top or back of box opposite the cover, if I extended a 12x12x6" JB would the 6x2" conduit go out of play and only focus on the 312.6 article at that point and the inches for bending radius be a factor? It's 3 phase with neutral 3/0 awg

The 6 X factor for box dimensions does out of play for a measurement when the conduit enters opposite the removable cover and 312.6 takes precedent. You would still need to maintain the 6 X between raceway entries fro conduits containing the same conductors.
