Does the MAX VOLUME inside an LL only apply if splicing conductors? I'm running a single 2.5" EMT from a distribution board to a panel, and the wire required is 4-4/0 and a #4G. My calcs bring the total cu in to 1.3772 and the max stamped is 3-250s (total cu in=157.3) Does this require me to upsize the LL to a 3.5"? I am not splicing.

IMO, yes you must upsize the LL
 
Your calculation should be based on 6X the trade side of the 2.5" EMT not the cubic inch area.

If you are on the 2017 edition, there is new language in 314.28(A)(3) to allow the equivalent for different conductor sizing and number than is marked. But I don't think in your case it will help you as (I didn't do the math) I think 4, 4/0 and 1 #4 is likely much more than 3, 250's.
Better order up that larger LL.
