On a 4” die cast Lb that has a rating of 3. 400 mcm conductors how would you calculate the exact mils allowed if using a different size conductor ?

Maximum of 1200 kcmils (3*400) using a combination of conductors smaller than 400 kcmil.

This generally all about the bending space of the conduit body more so than cross sectional fill. Cross sectional fill should be able to take anything the connecting raceways can handle. Had this application had all 12 AWG conductors you should be able to pull whatever max fill of the raceway is through the conduit body, but large conductors are limited by wire bending radius and what may potentially damage the conductor.
