I need to calculate the load for my single family dwelling.
are you only allowed to apply the demand factors from table 220.42 to either the neutral or the hot, but not to both, in the neutral and hot load calculation for my single family dwelling?

The service load calculation result is in volt-amperes, so it is not with respect to the "hot" or the neutral until you divide by volts to determine the minimum ampacity required. That said, dividing that result by the voltage is the minimum "hot". The neutral can have additional reduction of its minimum ampacity per 220.61, and that'll require separating line-to-line loads from line-to-neutral and multiwire branch circuits so you can determine the maximum unbalanced load on the neutral.
