I’m working on a project where I need to transition the wiring in a wall (single gang mud ring) to surface mount pipe. It could be considered a wet location so I can’t just use a single to 4sq extension box.

Can I make the transition by attaching a single gang bell box extension (w/ threaded hubs and a blank cover) to the single gang mud ring?

Since you mentioned a single gang mud ring, I'm assuming this is a commercial environment.
Since this is a commercial environment I'm assuming it's an existing pipe and wire project.
Assuming this is a pipe and wire project, I'm assuming single conductor Thhn/Thwn wet location rated conductors are present inside the flush mount box.

My 2 cents worth would be if you want to mount a bell box extension over the existing mud ring to convert to pipe, run a bead of silicone around the top and sides of it for good measure and go for it.
