For an ongoing project, we are sending 120VAC as discrete outputs from a PLC and currently have two wires to control the valve. Does NEC require a third grounding conductor as well? Could we use the conduit as the ground instead? Thanks

you can use metallic conduit as the egc.

Fitting or cord on valve.

Fitting or cord on valve.

If the valve has fittings no problem using the conduit. But if it has cord going to it I would use a ground wire because you would need to tie it to the conduit or pullbox and that is not always easy to find a spot or way to do it.
