Grounding wires do not carry current except in a fault situation. Do you mean grounded conductors (neutrals)?

No, and no, in that order. There may be an issue with conduit fill, since you have to count the EGC within the 40% limit. But you don't count them as current carrying conductors, for the purpose of derating.

One may be required depending on the conduit or the install.

I agree with what the others have said but would like to suggest reading 250.122(C).

310.15(B)(5) reads that grounding and bonding conductors are not required to be counted as current carrying conductors for ampacity derating purposes.
