For Class II, Division 2 areas, we have installations where the rated conduit runs vertically down and horizontal more than 5f (vertically) and 10ft (horizontally) but then runs vertical upwards before the the cable is moved onto tray (all within the div 2 area). Are conduit seals required for this given it has a vertically upward run or do the down/sideways rules apply.

As you have described it, you are fine.

It can be a bit difficult to explain, but the placement is always with respect to the protected enclosure. Any run 10' horizontal or 5' vertical downward from the raceway entrance to the enclosure is acceptable. Technically, as long as at least one 5' vertical downward run from the entrance occurs closest to the enclosure, other vertical runs may occur in other parts of the run.

Also note there is no difference for sealing requirements between Division 1 or Division 2 in Class II. Seals aren't required to be explosionproof in any case.

Okay that's great, thanks.
With the seal not required and rated conduit and fittings used (without a seal). There is the issue of the other end of the conduit being open when the cable is entering the tray.
This would still have an open path (although small) along the conduit into the instrument/equipment.
Is a gland required?

Sorry for the mundane questions, I've never worked with NEC, only IEC where we use armored cable with rated fitting and negate the requirement for conduit.

As long as the enclosure in question is properly "sealed", the open end can be filled with dust. It will need a bushing to avoid abrasion of the cables. For a warm, fuzzy feeling it may be packed with duct seal.
