Can you mix conductors from a 120/208 system with a 277/480 in the same J-box without any physical barriers?

Not if there are devices.

This graphic depict whats Derek mentioned. I believe that there is a scenario where electrically these two adjacent devices will have less than 300 volts between them. In a junction box the two voltage systems are permitted together. You just need to be aware of the rules regarding the identification of the grounded conductors.

If there are no devices in the box then look at 300.3(C).
