I have a 112.5kVA 3PH 4W 480V transformer located 3' from the edge of a sink, indoors, in a commercial space in the back-of-house in a restaurant space.
Is NEMA-2 enclosure sufficient, or it needs NEMA-3R?
I know that NEMA-2 protects against falling dirt and minor amounts of falling non-corrosive liquids/water in indoor environments.

I wouldn't worry about it at all. For most small dry-type transformers, like this example, the difference between an indoor and outdoor installation is the field addition of 'weathershields' which prevent rain from being blown into the enclosure.
