Is it required to run a neutral conductor to a disconnect switch that is powering a transformer? Primary voltage is 208, secondary voltage is 480. The feed is coming from a distribution panel and powering an elevator. It is 3 phase.

What type of step-up transformer are you using a Delta-Wye? If so there would be no place to connect the primary neural in the transformer. If it were Wye-Delta you would have an X0 on the primary but you would not connect a neutral to it.

Nor a ground. Either one would allow disastrous circulating currents under some conditions.

You post seems to imply that your using a standard 480 delta primary to 208Y secondary transformer in reverse. If so, this will present significant other issues that may not be suitable for an elevator. You should be using a 208 delta X 480Y transformer.
