Note on a drawing for a commercial project calls for 400amp rated conductors in a 3" grc (single phase) for the service drop, which according the 310.16 makes me think 600mcm. But then a note from the CT cabinet of the service to the first panel calls for 500mcm in a 3" grc. Think I would be safe with 500mcm for the service drop?

I think it's RFI time... I just think it's a simple typing error on the drawing.

Whats the calculated load? Less than 380A?

Looks to me like the shown calculated load is about 240amps but there are several "existing loads" shown. I think for the purposed of bidding I will price out for 500mcm.

Then I agree with Jude, typo, left out the 500's on the riser.
