Does a 240 single phase pump, connected by twist lock plug, not used for a pool need GFCI protection? I'm sure it does not however someone told me today that it's required by the NEC. The pump is for a system to spray calcium cloride onto salt in a dump truck.

I do not see any rule that requires it unless there is a manufacturer requirement. GFCI is not required for 240V rec., excluding pools- maybe something else but in general- not required.

What is the amperage?

(C) GFCI Protection. Outlets supplying pool pump motors
connected to single-phase, 120 volt through 240 volt branch
circuits, rated 15 or 20 amperes, whether by receptacle or by
direct connection, shall be provided with ground-fault circuitinterrupter
protection for personnel.

OOPs. Not a pool.
