Would you consider a CB on a rooftop unit located behind a cover secured by two screws to be readily accessible?

No, I wouldn't. See the definition of "accessible, readily" in article 100.

I would, if the motor or device that is being disconnected is also behind that same cover.

Steve, I've checked the definition but question the intent of using the words "quickly" and "obstacles." Is removing two screws considered an obstacle, or are they referring to more traditional obstacles i.e. equipment? Can the screws be removed "quickly?" I know... subject to interpretation of AHJ.

What kind of rooftop unit are you talking about? I assumed you were talking about an A/C.

Sometimes "readily accessible" means from a specific starting point, for instance in the case of an air conditioner..

440.14 Location.

Disconnecting means shall be located within sight from and readily accessible from the air-conditioning or refrigerating equipment. The disconnecting means shall be permitted to be installed on or within the air-conditioning or refrigerating equipment.

Than my opinion is if the panel with two screws is a service panel of the A/C than you're fine.
