Per code single conductors placed in cable tray must be 1/0 or larger. What if you have a 120 volt control circuit with say, #12awg? Must this be multi conductor cable?

Control conductors of that size must be multiconductor. See California Electrical Code Section 392.3(B)(1) for permitted uses of single-conductors in industrial establishments.
