I have wiring going into sump pump located in tank concrete pad Class 1 Division 1 area. The sump pump is single phase 208V fed from 20A breaker has 2#12 conductors however their is no equipment grounding conductor provided. Does code require equipment grounding conductor for gas station sump pump?

Yes, an equipment ground is necessary, and if the conduit is all metal, then it can serve that purpose.

Just an aside, it's a sub pump (submerged), not sump pump. Just sayin. :)
