No it cannot be a branch circuit. Branch circuits cannot supply panels.

No, it cannot be classified as a service when it is supplied from another panel. A service can only be provided by the utility and that service stops at the first overcurrent device.

You told us you are running a line from a temporary service panel to a subpanel. That being the case it is a feeder you are running and feeders must include an EGC, in your case you must have 2 hots, a neutral and a EGC. Four wires total, one can be bare.
This is not a maybe :happyno: and there is no way to code compliantly run less wires. A few code cycles back you might have been able to use the neutral as the grounding conductor but that allowance is gone.
Are you also aware you must install a grounding electrode system at the new sub panel location?
