If the speaker wire is coming from the amplifier then into a conduit with 2 hot legs neutral and ground(240v) will I have any interference?

Hum is a real possibility, but you can't run speaker wiring in the same conduit with power wiring anyway.

See 640.9(C). If the output of the amplifier is a Class 2 or 3 circuit, that section will send you to 725.133, which sends you to 725.135, and thosei rules will likely prohibit the mixing of the power and amplifier cables in the same raceway.
