Feeding single phase 480V-120/240V transformer, grounding

I have a switchboard circuit feeding a single phase 480V-120/240V transformer (50kVA). Is a ground conductor suppose to be ran with the current carrying conductors? Or is it just two hot, L1-L2?

For the primary since it 1Ø you'll need only 480 volts with two ungrounded conductors. On the secondary you'll need a neutral conductor for 120 volts. Not sure what you mean by a "ground conductor".

Grounded, no; grounding, yes. You only need a neutral in a feeder that feeds loads needing one.

The primary circuit needs an EGC run with it. Potentially can be metal raceway as the EGC.

The derived secondary system needs a GEC run to the building GES.

It can be possible to have one conductor sized for the larger of the two that fulfils both purposes.
